Long-Read Sequencing

Long Read Sequencing (LRS) instruments can sequence DNA molecules greater than 1000 bp, unlike its NGS counterpart, with average read lengths between 10,000 and >100,000 bases. The Core offers three instruments for LRS; the Oxford Nanopore Technology MinIon and PromethIon, and the Pac-Bio Sequel IIe and Revio.

The MinIon can generate 10-30 Gb of data per flow cell. This instrument is available for users to train and use on-site for cost-effective sequencing. Its higher-throughput staff-run complement, the PromethIon, can generate 50-150 Gb of data and 250 million reads per flow cell.

The Pac-Bio Sequel IIe and Revio use highly accurate (>99%) single-molecule real-time (SMRT) technology. The Sequel IIe can deliver roughly 20Gb and  4 million reads per flow cell while the Revio can deliver roughly 60-90 Gb and 10M reads.

The Bauer Core accepts sequencing samples from our Harvard community and external clients.
